Background: TumorNecrosisFactorAlpha（TNFalpha）isaproteinsecretedbylipopolysaccharidestimulatedmacrophages,andcausestumornecrosiswheninjectedintotumourbearingmice.TNFalphaisbelievedtomediatepathogenicshockandtissueinjuryassociatedwithendotoxemia.TNFalphaexistsasamultimeroftwo,three,orfivenoncovalentlylinkedunits,butshowsasingle17kDabandfollowingSDSPAGEundernonreducingconditions.TNFalphaiscloselyrelatedtothe25kDaproteinTumourNecrosisFactorbeta（lymphotoxin）,sharingthesamereceptorsandcellularactions.TNFalphacausescytolysisorcytostasisofcertaintransformedcells,beingsynergisticwithinterferongammainitscytotoxicity.Althoughithaslittleeffectonmanyculturednormalhumancells,TNFalphaappearstobedirectlytoxictovascularendothelialcells.OtheractionsofTNFalphaincludestimulatinggrowthofhumanfibroblastsandothercelllines,activatingpolymorphonuclearneutrophilsandosteoclasts,andinductionofinterleukin1,prostaglandinE2andcollagenaseproduction.TNFalphaiscurrentlybeingevaluatedintreatmentofcertaincancersandAIDSRelatedComplex.
